I don't think they need to necessarily add toughness as much as they need to add mass. Since the Niners OL is built to be lighter, smaller and more athletic, they win on angles and movement. I think the perception that they lack toughness stems from their inability to line up against guys, drive them straight back and finish them. They don't get pancake blocks or bury guys into the turf because that's not how they're built; in pass pro, they struggle to anchor against power. It looks even worse when they're going against light boxes and getting dominated at the line of scrimmage.
What they are lacking is mass; size; strength; power. The ability to move bodies at will. The OL would seem much tougher if they were bulldozing guys on short yardage situations or brute forcing holes open against a defense's will; stonewalling bullrushers; piledriving defenders into the grass rather than just getting in-between them; etc.
